Are you using a token in From email, without any default email ID? If yes, please add a default email ID to fix this.
Marketo SFDC Sync can process up to 20k records per hour. As you said, the sales team is adding leads directly to the campaign, I don't think there would be any lag. It should not take more than 5-10 minutes.

Adding on to Greg's point, Edward Unthank at Etumos has a product which allows you to get only valid email IDs. You can try that.
You can use a Middleware like Josh mentioned. You can also use the webhook and API approach. However, the 2nd approach will not allow you to fetch all the details from MKTO.
Theoretically your approach should work. Though, I would recommend using 2 different smart campaigns. Flow for Campaign 1: Wait till January 04 --> Request Campaign 2 Flow for Campaign 2: Wait for 48 Hours --> Send Email
You can use MKTO API and build a function, which can pull data from Lead Activity for all the leads. Once you have this data, you can use Tableau or any other tool for reporting this data.
