For background - We have a large number of partitions (23+) and workspaces and we would like to make some adjustments to the assignment rules of partitions that are currently in use in our production environment. We have tested these changes in a sandbox environment and have mapped out our changes in advance of these updates.
How does Marketo process these changes, will it run all of our contacts through the new assignment rules? What sort of processing time should we expect for any downtime while the system processes and shuffles around our contacts? Is it recommended to turn off the SF sync during this process or can it/should it remain live?

That's correct, it's only on lead creation (from CRMs and API). Existing leads need Smart Campaigns to reassign their partitions based on changes/activities.

we never use assignment rules in the partition admin, for the very simple reason that it does not provide enough control.
We always create a dedicated program and smart campaigns in a workspace that can access all partitions.

Furthermore, partition rules set in the admin work only for lead creation, not for lead update.

That's a really interesting approach to address the limitations of the assignment rules. We have gone the other direction and pushed some of the logic upstream into our SF CRM by using formula based fields that we then operate off of in Marketo assignment rules.
Gregoire are you able to address any of the specific questions above, especially the one around does the system "reprocess" all of our contacts after a change?
I do not think is does reprocess all persons after a change, because the system PAR's are fired on lead creation only. But I do not know for sure, since we never use them
