I have a custom field on our SFDC Opportunity object that I need to use as a constraint in Marketo. I've made the field available (read only) to the Marketo user in Salesforce. It's been a little over an hour – how long should it take for that to show up as a constraint on the Marketo side (or is there anything else I need to do to make it visible)? Our sync is working successfully otherwise.

Normally, it should take only a few minutes, unless you have some sync backlog. Look into the admin -> salesforce and check it you have a sync that has started a long time ago.
If you are using a Pro edition of SFDC, you also need to have the field added to a page layout, oftherwise the field will not be available to any profile / user.

Thanks, Greg. That was my understanding, too.
I've kept an eye on the sync status and haven't seen anything out of the ordinary.
And the field is available on at least one page layout on the Salesforce side. Not sure if this helps, but when I look at the Salesforce support debugger on the Marketo side, the field is visible, but carries the notation: * field not in Marketo (either new in SFDC or excluded from sync)

make sure that field is visible to Marketo user and/or contact Support.
Following-up here in case it helps anyone else down the road. After contacting support, the constraint appeared as expected. Ultimately it took about three hours to show up (which, according to support, was "a bit longer than normal" but could have been due to a backlog).
So, patience.
