No, you do not have to test anything at all (nor can you make any modifications to Marketo's cert bundle that it uses to communicate w/SFDC). This is entirely in Marketo's wheelhouse.

Sharing a response I got from Kyle on Marketo Support:
Salesforce changing their SSL cert will not change the endpoint for the sync with Salesforce so there will be no interruption of service and nothing which you will need to do in Marketo. The update won't have an impact on your Marketo functionality.
