Yes, just use a "Not Opened Email" filter for your second campaign.
Now, remember that email open detection is not reliable as it is based on the download of a tracking 1pixel image. And many people do not download images when they open the emails (this is the default setting for Outlook). So you may end up with a wrong target in your campaign.
