If you're using Salesforce, the way to do this is to use the Owner fields in the signature - because presumably each lead is owned the appropriate rep already.
I am not using Salesforce.
We also do not have SFC, so created custom fields for the Account Owner, by name, email and phone. You can then set up tokens based on those fields. It works quite well if you have those populating correctly. You can also build the email token such that the mailto works to send to the correct person. If you need further details on how we set ours up, let me know.
